Standardised portions of buccal swabs were extracted using a modified forensicGEM extraction procedure. The extracts were then profiled to ensure that the DNA gave usable profiles and the correct alleles. The quality of the genetic profile obtained was assessed using four criteria: 1. total peak heights 2. sister peak height ratio 3. visual indication of degradation or inhibition and 4. presence of non-allelic peaks. Results showed no indication of degradation or inhibition, and no discordances were observed. This forensicGEM method provides DNA from buccal swab reference samples highly suitable for PCR STR amplification using industry-standard methods. It is quick, largely hands-free and allows high throughput processing manually or using automation workspaces.
