This application note reports on the use of prepGEM as a DNA extraction method applied to multiple storage methods, including Queen’s lysis buffer, FTA cards, ethanol, and frozen whole blood. DNA from avian blood samples was extracted from the four different storage methods. The DNA yield for each storage type was significantly higher than with mammalian blood (this is expected given that avian blood cells are nucleated; as a result, very small amounts of avian blood may be used for DNA analysis). Sequencing from the barcoding products resulted in high-quality sequences. This extraction method for avian blood is rapid, reproducible, and provides DNA that can be reliably used for sexing and generating high quality DNA sequences.
